Specific Activities of Antioxidases and Malondialdehyde Contents of Valeriana jatamansi Jones Leaves Under Shading and Open Field Cultivation

Specific Activities of Antioxidases and Malondialdehyde Contents of Valeriana jatamansi Jones Leaves Under Shading and Open Field Cultivation
This research aims to elucidate the differences of the enzymatic antioxidant abilities of Valeriana jatamansi Jones leaves under the conditions of shading and open field cultivation, and to provide a reference for studying the responses of V. jatamansi to sunny environments and formulating the supporting techniques of field cultivation of V. jatamansi. The V. jatamansi leaves under the conditions of the shading and open field in the central part of Yunnan Province were used as the test materials, and the soluble protein content and the specific activities of the antioxidases, i.e., catalase (CAT), peroxidase (POD) and superoxide dismutase (SOD), and the malondialdehyde (MDA) content of the leaves were determined with spectrophotometry. Compared with the shading condition, the open field condition decreased the soluble protein content of the V. jatamansi leaves by 11.30%, but increased the specific activities of the CAT, POD and SOD, and the MDA content of the leaves by 11.43%, 30.90%, 21.58% and 18.48%, respectively. The differences of the soluble protein content and CAT specific activities of the leaves under the condition of the shading and open field cultivation did not reach the significant level, whereas those of the specific activities of the POD and SOD and the MDA content all reached the significant level. Under the conditions of open field cultivation, the soluble protein contents of the V. jatamansi leaves declined and the peroxidation-induced damages of the cellular lipids became worse, but the specific activities of the CAT, POD and SOD rose simultaneously to strengthen the enzymatic antioxidant ability of the leaf cells.

Bioassay-guided separation by use of the fission yeast expressing NES of Rev, a HIV-1 viral regulatory protein, resulted in isolation of valtrate (1) as a new Rev-transport inhibitor from the nucleus to cytoplasm from Valerianae Radix. Valtrate (1) also inhibited the p-24 production of HIV-1 virus without showing any cytotoxicity against the host MT-4 cells.

Rational design by the MO calculation disclosed 5,6-dihydrovaltrate (2) as the bioisostere of valtrate (1), the Rev-export inhibitor with anti-HIV activity. The synthesis of 2 was accomplished by ingenious use of asymmetric Diels-Alder reaction and stereoselective epoxidation associated with the adjacent hydroxyl group. Because of similar biological potency to 1, the analog 2 should be recognized as a promising scaffold for new anti-HIV agents with an unprecedented mechanism of action, inhibition for nuclear export of Rev protein, in the conventional remedy.

Petiole expiants of Valeriana wallichii. DC., a threatened medicinal plant, were used for inducing callus. Optimum callus formation was observed on Murashige and Skoogs' (1962) medium supplemented with 3.0 mg/l NAA and 0.25 mg/l Kn. Shoot regeneration was achieved upon transferring the callus to medium containing 1.0 mg/l Kn and 0.25 mg/l NAA. Complete plantlets were obtained on the same medium or upon transfer of the regenerated shoot buds to medium containing 5.0 mg/l Kn and 1.0 mg/l IAA. Nearly a thousand callus regenerated plants were successfully transferred to the field following previously standardized hardening procedures.
