Abstract

In order to establish a tissue culture and rapid propagation system, taking stems with axillary buds of Lagerstroemia indica‘Xiaoming 1’as testing materials, basic medium, concentrations of plant growth regulators, proliferation and rooting stage, and the transplanting method were studied. The results showed that the medium DKWBA 1.0 mg/LNAA 0.05 mg/LPVP 300 mg/L was suitable for inducing germination of stem auxiliary buds, and the rate of germination was up to 95.56%. The best proliferation medium was DKWBA 1.0 mg/LIBA 0.05 mg/L, and the multiplication coefficient was 5.08 with strong plantlets. The rooting percentage, average root number and length per shoot reached 100%, 6.2 and 4.12 cm, respectively, on the medium 1/2DKWNAA 0.1 mg/LAC 300 mg/L. Taking nutrition pot as container, peat soil and perlite as transplanting medium (3:1 or 2:1), the transplant survival rate was over 95.33%. There were many lateral roots and the plantlets which grew rapidly and vigorously. This method was suitable for tissue culture of L. indica ‘Xiaoming 1’and laid foundations for its large-scaled propagation and large area promotion.
