Study on the Tissue Culture of Pinus bungeana
The aim was to establish efficient regeneration system. Mature embryo and cotyledon of Pinus bungeana were used as explants, after primary culture and propagation culture, the role of different plant growth regulator type and concentration, activated carbon, GA3 in the various stages of tissue culture were studied. The results showed that the best initiated medium for cotyledon was MS+6-BA 3 mg/L+NAA 0.1 mg/L+TDZ 0.01 mg/L, the best initiated medium for mature embryo was MS+6-BA 2 mg/L+NAA 0.1 mg/L. The optimum shoot proliferation medium was MS+NAA 0.05 mg/L. Pre-embryo (removal of hypocotyl) as explants, the optimum shoots rooting medium was 1/2MS+IBA 0.3 mg/L. Activated carbon could not promote shoot proliferation, growth coefficient decreases with the increase of carbon concentration. Right amount of activated carbon and GA3 effectively promoted the growth of buds.
