The Phylogenetic Relationships of Eight Acer Species Based on Evidences from ITS Sequences

Abstract

In this study, the sequences of nrDNA ITS from eight varieties of Acer species were analyzed by PCR amplification and sequencing. The sequences of PCR products were about 750 bp including the whole ITS 1, 5.8 s, ITS 2 sequence and partial 18 s, 26 s rRNA gene fragment. The length of ITS1 and ITS2 sequences were 221~236bp and 231~237 bp respectively. There were distinct variant sites between the ITS sequences. Phylogenetic analysis of eight varieties of Acer species was carried out on the basis of ITS and the homology relation among them was elucidated from the molecular level, which provide the basis for the selecting of rootstock in the grafting of Acer Platanoides ‘Royal Red’.
