AFLP Analysis of 7 SH Line Apple Rootstocks

Abstract

The improved method of CTAB-DNA isolation was used to extract genomic DNA from leaves of 7 SH lines (Malus domestica Borkh. X Malus. Honanensis Rehder), which are used as apple rootstocks. The materials were analyzed by AFLP (Amplified Fragment Length Polymorphism)-silver staining protocol in order to detect blood relationship and genetic diversity. 20 pairs primers were selected from 143 primer pairs(3+3)to amplify the genetic DNA. Total 458 distinct amplification bands were obtained, 261 of them were polymorphic markers and the polymorphic frequency was 57%. UPGMA cluster analysis based on simple matching coefficient (SM) divided the 7 SH line apple rootstocks into 2 groups at 0.58 level.
