
复合抗生素对生菜种子萌发及土壤酶活性的影响
Impact of Compound Antibiotics on Lettuce Seed Germination and Soil Enzyme Activity
为探讨抗生素和灌溉水源变化对生菜、土壤的影响,以及其在培养第3、7 d时间上的变化,设计添加11种常见抗生素的复合污染水平(0、2.2、5.5、11 mg/kg)和2种不同灌溉水源的种子萌发及胁迫试验,并在发芽期间收集2次样品。结果表明,大部分生菜种子萌发指标(如种子的发芽势、发芽率、鲜重和苗长)在处理组与对照组间没有显著差异。在添加11 mg/kg浓度抗生素且灌溉水源为再生水时生菜种子的根长被显著抑制。采样期内(培养的第3、7 d),抗生素的添加和灌溉水源的变化没有扰乱生菜种子的抗氧化系统正常工作。培养第3~7 d,大多数处理组的土壤脲酶、土壤碱性磷酸酶活性有所上升,部分处理组与对照组之间存在显著差异,土壤酶活性相比植物萌发指标和植物酶活性对抗生素的添加更为敏感。
To investigate the effects of antibiotics and changes in irrigation water source on lettuce, soil and their changes in time on the 3rd and 7th day of incubation, seed germination and stress tests were designed with the addition of 11 common antibiotics at compound contamination levels (0, 2.2, 5.5 and 11 mg/kg) and two different irrigation water sources, and samples were collected twice during germination. The results showed that most of the lettuce seed germination indices (e.g., seed germination potential, germination percentage, fresh weight, and seedling length) were not significantly different in the treatment group compared to the control group. Root length of lettuce seeds was significantly inhibited when the concentration of addition of antibiotics was 11 mg/kg and the source of irrigation water was reclaimed water. During the sampling period (3rd and 7th day of incubation), the addition of antibiotics and the change in irrigation water source did not disturb the normal functioning of the antioxidant system of lettuce seeds. Soil urease and soil alkaline phosphatase activities increased in most of the treatment groups from the 3rd to the 7th day of incubation, and there was a significant difference between some treatment groups and the control group. Soil enzyme activities were more sensitive to the addition of antibiotics than plant germination indexes and plant enzyme activities.

The residue of antibiotics is becoming an intractable environmental problem in many organic vegetable bases. However, their residual levels and distribution are still obscure. This work systematically analyzed the occurrence and migration of typical veterinary antibiotics in organic vegetable bases, northern China. The results showed that there was no obvious geographical difference in antibiotic distribution between soil and manure. A simple migration model can be easy and quick to predict the accumulation of antibiotics in soil. Antibiotics were mainly taken up through water transport and passive absorption in vegetables. The distribution of antibiotics in a plant was in the sequence leaf>stem>root, and performed biological accumulation. The residues of antibiotics in all samples in winter were significantly higher than those in summer. Overall, this work can lay the foundation for understanding ecological risk of antibiotics and their potential adverse effects on human health by food chain.Copyright (c) 2010 Elsevier Ltd. 为认识橡胶废水沼液中外源溶解性有机物(dissolved organic matter,DOM)对抗生素污染生物效应的影响,本文采用水培和土培两种方法研究土霉素(oxytetracycline,OTC)对小白菜种子的萌发效应,在此基础上探讨外源DOM输入后土霉素对小白菜发芽率和胚根、芽伸长的影响。结果表明:(1)水培中DOM促进种子萌发,土培中抑制萌发;土霉素在水培0~50 mg/L、土培0~100 mg/kg胁迫下小白菜种子发芽率影响差异不显著;(2)在水培中,外源土霉素浓度与小白菜的胚根、芽长抑制率之间均呈显著正相关性;在土培中,外源性土霉素浓度与小白菜的胚根、芽长抑制率呈显著负相关性;(3)在水培和土培中,与对照组相比,DOM对小白菜的胚根伸长和芽伸长均起促进作用,水培促进率最高达55.05%,土培促进率最高达27.46%;(4)水培条件下,土霉素与DOM共存时对小白菜胚根伸长的抑制作用更明显;土培条件下,土霉素与DOM共存时,低浓度的DOM促进小白菜的胚根伸长,随着浓度的升高,促进作用下降。

通过水培实验研究了在1,10和30 mg/L浓度下,金霉素(chlortetracycline, CTC)和其代谢物差向金霉素(4-epi-chlortetracycline, ECTC)胁迫对小白菜种子的萌发、幼苗的早期生长、幼苗中抗氧化酶系统活性的变化以及CTC和ECTC积累进行了研究。结果表明,根长较其他指标对CTC和ECTC生态毒性实验更为敏感,小白菜体内SOD、POD和CAT活性被CTC诱导,活性逐渐升高,其中SOD、POD活性均高于对照,低浓度ECTC促进了SOD、POD和CAT活性的升高,高浓度时POD和CAT活性被抑制,且随浓度升高,抑制作用加强,表明小白菜抗CTC胁迫的能力强于ECTC。CTC和ECTC能在小白菜地上部分积累,处理浓度越高,吸收和积累量越多,其中CTC被积累作用强于ECTC。

为了查明安新县地下水水化学特征以及水质现状,选取45组地下水,采用仪器分析和容量分析相结合的方式进行测定,综合运用描述性数理统计和三元相图,分析安新县地下水的主要离子特征,并运用综合评价法对地下水质量予以评价。结果表明:(1)该区域地下水偏弱碱性,水化学类型主要以HCO<sub>3</sub>-Na型为主。(2)pH和偏硅酸在空间上变异最低(CV<30%);K<sup>+</sup>和NO<sub>3</sub><sup>-</sup>的空间变异性最高(CV>100%);其余指标的空间变异中等(CV:30%~100%)。(3)影响本区地下水环境质量的指标主要为TDS、SO<sub>4</sub><sup>2-</sup>、F<sup>-</sup>和总硬度。(4)水质评价结果显示该区地下水水质总体状况较好,多为Ⅲ类水,研究成果对了解安新县地下水水质现状具有重要意义。

以小白菜为受试生物,研究了6种常用抗生素(四环素、土霉素、金霉素、强力霉素、氯霉素和磺胺二甲嘧啶)对小白菜种子萌发、根(芽)伸长、可溶性蛋白及抗氧化酶系统的毒理效应。结果表明,0.5~5 mg·L<sup>-1</sup>抗生素胁迫下,小白菜种子的发芽率无显著变化,随着抗生素暴露浓度的增加,小白菜种子的发芽率受到了显著抑制;抗生素暴露浓度与小白菜根(芽)伸长抑制率之间具有良好的剂量效应关系。由抗生素对小白菜根长的半抑制效应浓度(EC<sub>50</sub>)可知,6种抗生素对小白菜种子萌发的毒性大小依次为:磺胺二甲嘧啶>氯霉素>金霉素>四环素>强力霉素>土霉素。实验浓度范围内的抗生素对小白菜幼苗体内可溶性蛋白含量具有显著的诱导效应,且各处理组间差异显著。此外,抗生素的胁迫破坏了小白菜幼苗体内的抗氧化防御系统的平衡。

农田土壤抗生素累积产生的生态风险已成为目前国内外研究的热点,土壤酶活性的变化对于研究抗生素影响下的农田土壤生态系统养分循环,具有重要的生态学意义。分别以酸性、中性和碱性农田土壤为研究对象,添加畜禽养殖业中常用的3种抗生素--土霉素(Oxytetracycline,OTC)、恩诺沙星(Enrofloxacin,ENR)和磺胺二甲嘧啶(Sulfamethazine,SM2)进行处理,分析了土壤中脲酶、过氧化氢酶和磷酸酶对抗生素的敏感性。结果表明,在添加抗生素处理的0-28 d过程中,脲酶活性整体呈现抑制趋势,过氧化氢酶活性整体呈现促进趋势,磷酸酶活性随着添加抗生素的不同呈现不同的变化趋势。在28 d时,与对照组相比,脲酶活性在添加3种抗生素的酸性、中性土壤中及添加ENR与SM2的碱性土壤中均显著受到抑制,抑制率为38.03%-96.08%,而在添加OTC的碱性土壤中受到促进,促进率为28.43%;过氧化氢酶活性在添加3种抗生素的酸性土壤中及添加OTC与SM2的中性、碱性土壤中均受到促进,促进率为24.88%-268.25%,而在添加ENR的中性、碱性土壤中受到抑制,抑制率为18.42%和29.04%;磷酸酶活性在添加OTC的3种土壤中均受到促进,促进率为14.73%-101.26%,而在添加ENR的3种土壤中均无显著变化,在添加SM2的3种土壤中均受到抑制,抑制率为61.83%-70.54%。该研究表明抗生素对土壤酶活性影响复杂,尤其是对脲酶活性的抑制,减弱了土壤氮循环相关酶的代谢功能及其稳定性,进而影响土壤生态系统的功能稳定性。

To evaluate the ecotoxicological potential of ten prescription drugs against aquatic organisms from different taxonornical classes, a set of biotests were performed using the cladoceran Daphnia magna, the chlorophyte Desmodesmus subspicatus and the macrophyte Lemna minor. Endpoints were immobilisation for Daphnia and inhibition of the average growth rate for Desmodesmus and Lemna. For most of the substances, toxicities were moderate, with EC(50)s in the range from 10 to 100 mgl(-1) or even far above, whereas Lemna was the most sensitive test species in the majority of all tested compounds. Tests with combinations of various pharmaceuticals revealed stronger effects than expected from the effects measured singly. Clofibrinic acid and Carbamazepine have been found to act by a non-specific mode of action (non-polar narcosis), and with Daphnia the combination effect of these substances followed the concept of concentration addition, while in the algae test the concept of independent action could be used to calculate the mixture toxicity. The anti-inflammatory drugs Diclofenac and Ibuprofen have also been found to act unspecific by non-polar narcosis and to follow the concept of concentration addition in the algal test as well as in the Daphnia test. The measured toxicities of the tested pharmaceuticals shows that acute effect of single substances in the aquatic environment are very unlikely. But we should keep in mind that considerable combination effects can occur and that toxicity data from chronic studies are needed to assess the environmental risk of drug residues.

采用盆栽试验研究了土壤外源四环素与金霉素(TC-CTC)、四环素与磺胺嘧啶(TC-SD)二元复合污染对小白菜幼苗生长发育的毒理效应。结果表明,0.5 mg·kg<sup>-1</sup>抗生素复合污染胁迫促进了小白菜芽长和根长的生长,但随着抗生素浓度的增加,小白菜芽长及根长生长逐渐受到抑制,根长受抑制程度高于芽长,25 mg·kg<sup>-1</sup> TC-CTC处理对根长的抑制率达72.7%。与抗生素单一污染相比,二元复合污染胁迫使小白菜体内可溶性蛋白含量显著降低,暴露30 d时25 mg·kg<sup>-1</sup> TC-CTC处理对可溶性蛋白抑制程度最大,抑制率为67.1%,同时超氧化物歧化酶、过氧化物酶和过氧化氢酶活性受到的抑制作用更显著,呈现协同作用。
