芫荽种质资源分子身份证构建

摘要

利用RAPD分子标记,对96份引进芫荽种质进行群体结构性研究并构建其分子身份证。结果显示,从300条引物中筛选出的23条扩增性强、清晰度高、稳定性好的引物,在芫荽群体中共检测到34个多态性条带,每个引物检测到等位基因分布在2~10之间,平均为4.61个。品种间特异性指数差异较大,种质特异指数介于69.964~208.694间,平均为104.29。在研究多样性基础上,利用S2025、S2007、S2020、 S2029、 S177、 S183、 S509、S169、S1260、S167、 S45、S24、 S43、 S29共14条引物构建了96份种质资源的分子身份证,可以有效区分各材料。

Abstract

The paper aims to understand the genetic information of the introduced germplasm resources, promote efficient distinction and rational utilization. Population structure analysis and molecular ID construction of 96 coriander samples were studied using RAPD. 23 from 300 random primers showed high polymorphism, 34 polymorphic bands were detected, and the quantity of detected alleles of the 23 primers ranged from 2 to 10, at an average of 4.61. The difference of specialty indices among varieties was relatively large, which ranged from 69.964 to 208.694, at an average of 104.29. Based on the diversity analysis, the molecular identities of the 96 coriander varieties were constructed by 14 primers: S2025, S2007, S2020, S2029, S177, S183, S509, S169, S1260, S167, S45, S24, S43 and S29. They can differentiate the samples effectively.
