茶树菇超高压保鲜过程中一株类芽孢杆菌的鉴定及系统发育分析
Identification and Phylogenetic Analysis of a Paenibacillus Strain from the Package of Agrocybe cylindracea Processed by Super High Pressure Technology
对超高压保鲜过程中茶树菇的包装袋分离到的菌株WM1003进行鉴定及系统发育分析,以菌株WM1003的基因组为模板扩增获得了该菌株的16S rRNA基因序列,经过序列分析及系统发育分析,对该菌株进行鉴定;PCR扩增结果表明,该菌株的16S rRNA基因的大小约为1600 bp,其GenBank登录号为HM748831;通过在线BLAST比对,结果表明该菌株属于芽孢杆菌属;基于16S rRNA基因序列的系统发育分析结果表明该菌株同已知菌Paenibacillus jamilae CECT 5266的相似性最高,为99.5%,初步将该分离菌株WM1003鉴定为Paenibacillus sp.;本文通过16S rRNA序列及系统发育分析,对菌株WM1003进行了鉴定,为茶树菇的超高压保鲜技术提供了理论支持。
The aim of this study is to identify the bacteria strain numbered as WM1003 isolated from the package of the Agrocybe cylindracea processed by super high pressure technology. The genomic DNA of the strain WM1003 was isolated and purified. 16S rRNA gene sequence was amplified using genomic DNA of the strain WM1003 as template and was sequenced. Phylogenetic position of the WM1003 was analyzed by using MEGA4.0 based on the 16S rRNA sequences. PCR product of the strain WM1003 was about 1600 bp. The 16S rRNA gene sequence of the strain WM1003 was accepted by GenBank as HM748831. Phylogenetic analysis based on the 16S rRNA sequences showed that the strain WM1003 held highest similarity with Paenibacillus jamilae CECT 5266 (99.5%). In the present study, the strain WM1003 was identified through 16S rRNA gene sequence and phylogenetic analysis. This laid the foundation for the application of super high pressurs technology in Agrocybe cylindracea.
