HRM及其在植物育种中的应用展望
Application of High Resolution Melting Analysis in Plant Breeding
HRM(high resolution melting,高分辨率熔解曲线)分析基于监测双链DNA升温变性解链的熔解曲线,是新发展起的一种基因突变扫描及SNPs(single nucleotide polymorphisms,单核苷酸多态性)鉴定技术。由于在灵敏度和准确性上具有突出的优点,HRM近几年来在突变扫描、基因分型、DNA甲基化研究中得到广泛应用。本文简述了HRM的原理及特点,同时结合HRM分析植物群体的实例,对HRM在植物育种中的应用进行了展望。
HRM (high resolution melting analysis), which based on monitoring melting (dissociation) curves of double-strand DNA, is a recent advance for the detection of mutations and SNPs (single nucleotide polymorphisms). Due to its outstanding performance at sensitivity and accuracy, HRM has been largely used for mutation scanning, genotyping and DNA methylation research in recent years. In this paper, the mechanism and feature of HRM were introduced briefly; furthermore, the prospects of its application in plant breeding were provided by combining with the cases of HRM in analyzing plant populations.
