SB-431542对梅花鹿鹿茸软骨层细胞增殖的影响
Effects of SB-431542 on the Proliferation of Sika Deer Antler Chondrocytes
通过研究转化生长因子β(transforming growth factor-β, TGF-β)的特异性小分子拮抗剂SB-431542对梅花鹿鹿茸软骨层细胞增殖的影响,探讨转化生长因子β在鹿茸快速生长中的调节机制。分离培养生长30天的梅花鹿鹿茸软骨层细胞,将传代培养第2代的细胞经不同浓度的SB-431542 (0,1,3,5,8,10μM)作用,培养48小时后用MTT法测其细胞增殖的变化,并用SPSS软件分析其差异性。结果显示,SB-431542处理组细胞增殖活性都低于对照组(P<0.05),其中浓度为8μM和10μM的SB-431542处理组与对照组相比差异非常显著(P<0.01),随着添加SB-431542浓度的升高,对细胞的生长速度抑制更加明显,试验表明,TGF-β对维持梅花鹿鹿茸软骨层细胞的快速增殖有重要作用。
The regulatory mechanism of TGF-β on the rapid growth of antler was investigated by studying the effect of specific micro-molecule antagonist SB-431542 of transforming growth factor- β (TGF-β) on the cell proliferation of sika antler chondrocytes. The sika antler chondrocytes were isolated after cultured for 30 days, then the second generation cells were incubated with 0, 1, 3, 5, 8 and 10 μM of SB-431542, after 48 hours of culture, the MTT assay was used to examine the proliferation of antler chondrocytes and the SPSS software was used to analyze the difference of the results. The result showed that the proliferation activity of cells treated with SB-431542 was lower than that of the control group (P<0.05), among them, the proliferation activity of cells treated with SB-431542 at the concentration of 8μM and 10μM was significantly lower than that of the control (P<0.01), with the increased concentration of SB-431542, the inhibition to cell proliferation rate was more significant. In conclusion, these results indicated that TGF-β might play an important role to maintain the rapid proliferation of sika antler chondrocytes.
