摘要

对已有方法和规范中样品前处理方法和色谱条件进行了优化和对比研究,建立检测虫草制品中腺苷和虫草素含量的反相高效液相色谱方法。样品经水超声提取40 min,离心过滤后进行分析。采用C18色谱柱(250 mm×4.6 mm,5 μm),以水-甲醇(85:15,V/V)为流动相,流速为1.0 mL/min,检测波长为260 nm。本方法检测限为0.2 μg/mL,定量限为1.0 μg/mL。以所建立的方法对虫草制品进行分析,样品加标回收率在94%~103%,相对标准偏差均小于3%。该检测方法简便、准确,并能同时测定腺苷和虫草素。

Abstract

To establish the quantitative analysis method for adenosine and cordycepin in Cordyceps sinensis by HPLC. The sample was extracted with pure water under ultrasonic for 40 min. After centrifugation and filtration, the extract was analyzed by HPLC with C18 column at 1.0 mL/min and detected at 260 nm wavelength. The mobile phase was composed of pure water and methanol(85:15, V/V) and the column temperature was 30℃ . This method showed the LOD was 0.2 μg/mL, LOQ was 1.0 μg/mL. The average recoveries of adenosine and cordycepin were 94%- 103% and RSD bellow 3% . The method had high sensitivity, selectivity and simple operation, and can be used for testing the adenosine and cordycepin.
