Fluorescein isothiocyanate (FITC), which was not toxic to protoplasts, was used for the fusion of protoplasts. After the fusion of the FITC-labelled monokaryon protoplasts from Pleurotus sapidus and the non-FITC-labelled monokaryon protoplasts from Pleurotus ostreatus, a pair of sticked protoplasts was obtained with micromanipulator under an inverted microscope. Cultured with liquid regeneration medium, the fusants were selected according to the clamp connection of dikaryocyte with a fusion rate of 5.2?0~(3). It was found that the estcrase isozyme zymogram of fusants changed greatly, while the biomass and growth rate of many fusants were superior to those of their parents, and that the fruitbody morphology of the fusants was different from their parents.
