Molecular Cloning and Expression Analysis of GhPDS1 Gene in Gossypium hirsutum

Abstract

Phytoene desaturase(PDS) play an important role in the physiological metabolism of carotene and virus induced silencing of foreign genes. The degenerated primers were designed from the conserved motifs of phytoene desaturase gene isolated from other plants in NCBI, and the middle fragments of PDS gene in Gossypium hirsutum were amplified. The 5’and 3’ends of the cDNA were cloned through RACE methods. The gene of PDS in Gossypium hirsutum was named as GhPDS1 (GenBank accession number: HQ441484). The semi-quantitative PCR results revealed that the transcript of GhPDS1 gene was the highest in flowers and then leaves. Phytoene desaturase gene is a endogenous gene as a visible marker to study the function of gene by endogenous gene silencing which causes cotton appear the yellow vein mosaic like pattern.
