摘要

本文对美味牛肝菌(Boletus edulis)液体发酵生产粗多糖的培养基和发酵条件进行了筛选。在实验范围内,美味牛肝菌发酵生产粗多糖的最适培养基配方为:马铃薯10.0%,蔗糖2.0%,蛋白胨0.6%,KH2PO40.3%,MgSO4.7H2O0.2%,CaCO30.2%,ZnSO4.7H2O0.1%。在初始pH5.0~5.4,振荡转速120r/min,培养温度25~28℃,250mL三角瓶中装液量40%,加10颗玻璃珠,发酵时间5d时摇瓶培养粗多糖产量最高,达3.161g/L。经10L机械搅拌发酵罐放大动态实验,研究pH值、溶氧、菌丝生物量(湿重)以及粗多糖产量的变化,可知最适发酵时间为96h。

Abstract

The effects of selected culture parameters on polysaccharide production by a strain of Boletus edulis are described.Optimal conditions for small-scale polysaccharide production in submerged flask cultures were as follows:a fermentation medium consisting of 10% potato,2% sucrose,0.6% peptone,0.10% ZnSO4·7H2O,0.2% CaCO3,0.3% KH2PO4 and 0.2% MgSO4·7H2O,pH 5.0~5.4,120 r/min shaking speed,temperature 25~28 ℃,40% medium loading ratio,10 glass beads per flask,and a 5-day incubation period.Under these conditions,polysaccharide production reached a peak of 3.161 g/L.In 10-L fermenter cultures using the same medium,pH,temperature,a 50% medium loading ratio,a similar stirrer speed,and an aeration rate of 0.8 vvm,highest polysaccharide yields were obtained after four days.
